EDIT - you can probably drop either Make the Call or Call for Backup to make the deck 40 cards.
Stay in AE for a few turns and control the early game with The Pericles if the Villian isn't Steady/Stalwart. If they are, fish for The Circe and jam Nick Fury asap.
Buff him up with Sidekick, Inspired and Sidearm, then use Command Team, Side-by-Side and Get Ready to continously ready him and Field Agent to protect him from damage.
This will allow you to put tokens on your S.H.I.E.L.D Supports, keeping them in play longer, whilst pumping damage and Thwarting with Nick.
Agent 13, Victoria Hand and On The Double can ready your Supports, allowing Nick or The Iliad more time to finish the Villian off.
